ABTS diammonium salt is a substrate for horseradish peroxidase (HRP) conjugate.

研究了使用 ABTS 作为 HRP 偶联物底物的酶联免疫吸附测定 (ELISA) 的显微技术。在 4 种底物的比较研究中，即；5-氨基水杨酸 (5AS)、邻苯二胺 (OPD)、邻甲苯胺 (OT) 和ABTS，对于HRP的灵敏度来说，ABTS是最灵敏、最稳定、目测最好的，呈蓝绿色^[1]。ABTS 是一种典型的过氧化物酶底物。为了纯化和表征，在培养上清液中产生活性蛋白的条件下，大规模 (XL) 培养过氧化物酶阳性转化体。培养 160 小时后，相对于底物 ABTS 的活性达到 55,000 U/L，收集含有过氧化物酶的上清液。以ABTS为底物，当 H₂O₂ 浓度升至 0.125 mM 以上时，过氧化物酶活性显著下降，表明该酶被 H₂O₂ 抑制。根据所测底物的不同，最大反应速率可达 31.2 至 125 μM^[2]。
