PKH 26 is a red fluorescent dye, PKH 26 can stably bind to the lipid region of cell membrane and emit red fluorescence (Ex/Em=551/567 nm), which is mainly used for in vitro cell labeling, in vitro cell proliferation studies and in vivo and in vitro cell tracing studies^[1].

一般方案
1. PKH 26 工作液的制备
1.1 储备液的制备
将 1 mg PKH 26 溶解在 1 mL DMSO 中以获得 1 mM 储备液
注意：建议储备液在 -20°C和 -80°C避光保存，避免反复冻融。
1.2 PKH 26 工作液的配制
用无血清细胞培养基或PBS按1:50或1:100稀释储备液，得到5-10 μM的工作液。
注意：请根据需要调整 PKH 26 工作液的浓度
2.细胞染色
2.1 悬浮细胞 (6孔板)
a.4℃1000g离心3-5分钟，弃上清. PBS洗2次，每次5分钟。细胞密度为1×10⁶/mL。
b.加入1 mL工作液，室温孵育10- 45分钟。
c.400g 4℃离心3-4分钟，弃上清。
d.PBS洗涤2次，每次5分钟。
e.用无血清细胞培养基或 PBS 重悬细胞。荧光显微镜或流式细胞术观察。
2.2 贴壁细胞
a．在无菌盖玻片上培养贴壁细胞。
b.从培养基上取下盖玻片并吸出多余的培养基。
c.加入100 μL工作液，轻摇使之完全覆盖细胞，室温孵育5-30分钟。
d．用培养基洗两次，每次5分钟。荧光显微镜或流式细胞术观察。

储存
